Interactions of human intestinal mast cells with bacteria and bacterial products

Abstract:

In our project we will analyse the interactions of human intestinal mast cells (hiMCs) with bacteria and bacterial products. hiMCs are in close proximity to the bacterial flora, they express numerous Toll-like receptors (TLRs), but surprisingly they are hyporesponsive towards bacteria under normal conditions. Therefore, we aim to analyse the mechanisms leading to the hyporeactivity of hiMCs towards bacterial products and TLR ligands. In detail, we will examine the role of NOD2 and other intracellular or extracellular molecules in bacterial sensing and mediating hyporesponsiveness in hiMCs. Second aim is to define changes in interaction between hiMCs and bacteria in gastrointestinal disorders such as Crohn´s disease or ulcerative colitis. Moreover, the mechanisms of hiMC activation by bacterial haemolysin will be unravelled. Third, we will evaluate if selected probiotic bacteria can modulate MC-bacteria interactions, or even balance altered MC reactions towards bacteria. The main goal of our proposal is to understand better the MC-bacteria-interactions at the gastrointestinal barrier, which may provide a basis for new and innovative therapeutic intervention
